چکیده

The oxygen evolving complex (OEC) of photosystem II (PSII) requires chloride as a critical cofactor. The Cl content of PSII was determined to be 1 Cl/PSII by the AgCl colloid assay. The dissociation constant for Cl in NaCl-washed PSII was determined using the Cl sensitive microelectrode. Two classes of Cl binding sites were found with Kd values of 53 μM and 4.2 mM. The effects of anions on the S2’ oxidation state of the OEC were investigated using electron paramagnetic resonance spectroscopy. It was found that chloride was

The mechanism of amine inhibition of the photosynthetic oxygen evolving complex Amines displace functional chloride from a ligand site on manganese

Amines inhibit photosynthetic oxygen evolution by binding to manganese in the oxygen evolving complex. This inhibition is more effective if chloride is absent. Steady-state kinetic analyses show that ammonia and chloride compete for the same site in the oxygen evolving complex; inhibition of oxygen evolution by either chloride depletion or by ammonia addition induces Signal IIr, which saturates...

Location of chloride and its possible functions in oxygen-evolving photosystem II revealed by X-ray crystallography.

The chloride ion, Cl(-), is an essential cofactor for oxygen evolution of photosystem II (PSII) and is closely associated with the Mn(4)Ca cluster. Its detailed location and function have not been identified, however. We substituted Cl(-) with a bromide ion (Br(-)) or an iodide ion (I(-)) in PSII and analyzed the crystal structures of PSII with Br(-) and I(-) substitutions. Substitution of Cl(-...

Towards models of the oxygen-evolving complex (OEC) of photosystem II: a Mn4Ca cluster of relevance to low oxidation states of the OEC.

Synthetic access has been achieved into high oxidation state Mn/Ca chemistry with the 4 : 1 Mn : Ca stoichiometry of the oxygen-evolving complex (OEC) of plants and cyanobacteria; the anion of (Et(3)NH)(2)[Mn(III)(4)Ca(O(2)CPh)(4)(shi)(4)] has a square pyramidal metal topology and an S = 0 ground state.

Effects of flashing light and hydroxylamine on the binding affinity of H C03- to photosystem II in chloroplast thylakoids

The affinity of HI4 CO, for an anion binding site on photosystem II in chloroplasts shows a period-of-two oscillation with flash number when formate is included in the reaction mixture. Although this behavior was originally explained (Stemler, A., 1979, Biochimica et Biophysica Acta 545, 36-45) as reflecting the oxidation state of the secondary photosystem II acceptor, B, we report additional r...

Minireview Photoinhibition – a historical perspective ∗

Photoinhibition is a state of physiological stress that occurs in all oxygen evolving photosynthetic organisms exposed to light. The primary damage occurs within the reaction center of Photosystem II (PS II). While irreversible photoinduced damage to PS II occurs at all light intensities, the efficiency of photosynthetic electron transfer decreases markedly only when the rate of damage exceeds ...
